Oomycete transformation

Transformation is an essential step in the functional analysis of proteins.  Here we show a GFP tagged membrane transporter expressed in P. sojae. The application of CRISPR technology in oomycetes will enable us to initiate targeted knockouts of key genes in oomycetes.

In this assay, we have used a multichannel pipette to array different Pseudomonas isolates on a V8 agar plate.  Then hyphal plugs of th pathogen Pythium heterothalicum were arrayed as hyphal plugs onto the plate. Inhibition by the bacteria is seen as a clearing zone around the bacterial colony.
